fa مطالعه مقدماتی آثار سیتوپاتیک ساب‌تایپ‌های 3-1 و 6 بلاستوسیستیس در رده سلولی HT-29 The Study of the Cytopathic Effects of Blastocystis sp., ST1-3 and 6 Isolated from Subjects Referred to Medical Laboratories in HT29 Cell Line انگل شناسی Parasitology پژوهشی Original <div style="text-align: justify;">سابقه و هدف: بلاستوسیستیس در حال حاضر یکی از شایع‌ترین انگل‌های تک یاخته‌ای روده‌ای است که در مدفوع انسان و حیوان یافت میشود. بیماری‌زایی بلاستوسیستیس و ساب تایپ‌های آن روی سلول رده گوارشی مشخص نیست. این مطالعه با هدف بررسی تفاوت بین ساب‌تایپی در القای آپوپتوز سلولی در رده سلولی HT-29 در سال 1399 و در پژوهشکده بیماری‌های گوارش و کبد دانشگاه علوم پزشکی شهید بهشتی انجام شد. روش کار: این بررسی یک مطالعه تجربی روی هفت ایزوله شامل شش ایزوله از ساب تایپ های 1 تا 3 و 6 از 30 ایزوله بلاستوسیستیس جدا شده از افراد دارای علایم بالینی و ساب تایپهای 1 تا 3 از 25 ایزوله بلاستوسیستیس جدا شده از حاملان بدون علامت بود. پس از <a name="_Hlk89257810">خالص‌سازی </a>و شمارش میکروارگانیسمها، آنتی‌ژن محلول بلاستوسیستیس از 105×1 میکروارگانیسم با استفاده از سه چرخه انجماد و ذوب در نیتروژن مایع و حمام آب 37 درجه سانتی‌گراد تهیه شد. سپس آنتی‌ژن محلول ساب تایپ‌های بلاستوسیستیس با رده سلولی HT-29 به مدت 48 ساعت انکوبه شدند. در نهایت، سرعت تکثیر سلولی با استفاده از روش MTT اندازه‌گیری شد. برای بررسی تفاوت‌های آماری در تکثیر سلولی بر اساس زیرگروهها و همچنین  علایم، از آزمون ANOVA یک طرفه همراه با GraphPad Prism استفاده شد. یافته‌ها: نتایج به وضوح نشان داد که تمامی ساب تایپ‌های بلاستوسیستیس در مقایسه با گروه شاهد تیمار نشده (NTC) دارای اثر سیتوپاتیک/ بازدارنده بر رشد سلول‌های HT-29 بودند. بیشترین میزان اثر ستوپاتیک در ساب تایپ 1جدا شده از افراد بدون علامت (25درصد) و کمترین آثار سایتوپاتیک در ساب تایپ 1 جدا شده از افراد با علامت بالینی (17/11درصد) دیده شد.  با این حال، هیچ ارتباط آماری بین ساب تایپ‌ها و همچنین علایم بالینی با اثر سیتوپاتیک وجود نداشت (0/11P-value =). نتیجه‌گیری: یافته‌های مطالعه حاضر نشان داد که اگرچه از نظر آماری ارتباط معنا‌داری بین ساب تایپ‌ها و اثر سیتوپاتیک وجود نداشت، اما به نظر میرسد که ویژگی های ایمونولوژیک میزبان نقش اساسی در پاسخ به بلاستوسیستیس دارد. در واقع، مقایسه بین ساب تایپ‌ها نشان داد که ساب تایپ 2 آثار خفیف‌تری را القا می‌کند. با این حال، بیماری‌زایی بلاستوسیستیس و تفاوت بین ساب تایپ‌ها بحث برانگیز باقی مانده است و نیاز به بررسیهای مقایسهای بیشتری دارد.</div> <div style="text-align: justify;">Background and Aim: Blastocystis sp., is one of the most common intestinal protozoan parasites found in both human and animal fecal samples. The pathogenicity and cytopathic effects of subtypes ST1-3 and 6 of Blastocystis sp., on gastrointestinal cell line are unclear. This study aimed to investigate the impacts of intra- subtype variations on the apoptosis and cytopathic effects on the HT-29 cell line in 2019 at the Research Institute for Gastroenterology and Liver Diseases, Shahid Beheshti University of Medical Sciences, Tehran, Iran. Methods: This study was an experimental investigation of Blastocystis sp., subtypes 1-3 and 6, which were isolated from stool samples of 30 symptomatic and subtypes 1-3 from 25 asymptomatic subjects. After purifying and counting the microorganisms, lysate of Blastocystis sp., was prepared from 1 × 105 of the microorganism by three freeze- thaw cycles in liquid nitrogen and a 37°C water bath. Then, the lysates of subtypes of Blastocystis were co-incubated with the HT-29 cell line for 48h. Finally, the cell proliferation rate was measured using an MTT dye assay. For measuring the statistical differences in the cell proliferation based on subtypes as well as symptoms, One- way ANOVA test incorporated with GraphPad Prism was applied. Results: The results clearly indicated that all Blastocystis sp., subtypes, compared to non-treated control (NTC), had a cytopathic/inhibitory effect on HT-29 cells growth. The lowest cytopathic effect was seen in ST1 isolated from symptomatic patients (17.11%), while the highest was seen in ST1 isolated from asymptomatic patients (25%). Indeed, ST2 showed milder cytopathic effects in the HT-29 cell line compared to ST1, 3, and 6. However, there was no statistical correlation between subtypes as well as symptoms and cytopathic effect. Conclusion: Although there was no statistically significant correlation between subtypes and the cytopathic effect, immunologic responses of hosts seem to play a critical role against Blastocystis sp.
