fa ارزیابی ژن‌هایTENT4A وTENT4B در سرطان کولورکتال: تحلیل مسیرهای سیگنالینگ و بررسی بیان در رده‌های سلولی Evaluation of TENT4A and TENT4B Genes in Colorectal Cancer: Signaling Pathway Analysis and Expression Evaluation in Cell Lines علوم سلولی( سلولی مولکولی، سلول های بنیادی) Cellular Sciences (Molecular Cells, Stem Cells) پژوهشی Original <div style="text-align: justify;">سابقه و هدف: سرطان کولورکتال (CRC) یکی از شایع‌ترین بدخیمی‌ها در جهان است و شناسایی نشانگرهای زیستی مولکولی جدید برای تشخیص زودهنگام و پیش‌بینی سیر بیماری از اهمیت بالایی برخوردار است. ژن‌هایTENT4A  وTENT4B  از خانواده پلی(A)  پلیمرازهای غیرکانونیک هستند که در تنظیم پایداری RNA و کنترل ترجمه نقش دارند. هدف این مطالعه بررسی الگوی بیان این ژن‌ها و تحلیل مسیرهای سیگنالینگ مرتبط با آن‌ها در سرطان کولورکتال بود. روش کار: این پژوهش یک مطالعه توصیفی بود. داده‌های RNA-Seq بیماران مبتلا به سرطان کولورکتال از پایگاه The Cancer Genome Atlas (TCGA) استخراج شد. ژن‌های دارای بیان تفاضلی (Differentially Expressed Genes; DEGs)، یعنی ژن‌هایی که اختلاف معنی‌دار بیان بین نمونه‌های توموری و طبیعی داشتند، با استفاده از بستهedgeR  شناسایی شدند. سپس تحلیل غنی‌سازی مسیرهای زیستی با استفاده از پایگاه‌های KEGG و MSigDB انجام شد. ارتباط بیان ژن‌ها با بقای بیماران و ارزش تشخیصی آن‌ها به‌ترتیب با آزمون Kaplan- Meier، رگرسیون کاکس و منحنی ROC بررسی شد. همچنین، سطح بیان ژن‌های TENT4A وTENT4B  در رده‌های سلولی HT29 و HCT116 نسبت به سلول HEK293 با روش Real-Time PCR ارزیابی شد. یافته‌ها: در مجموع 4558 ژن دارای بیان تفاضلی شناسایی شدند  .(FDR<0.05)ژن‌هایTENT4A  وTENT4B  افزایش بیان معنی‌داری نشان دادند، در حالی که ژن‌های TENT5A،TENT5B  وTENT5C  به‌طور معنی‌داری کاهش یافته بود(FDR<0.05). تحلیل غنی‌سازی، ارتباط معنی‌دار این ژن‌ها را با مسیرRNA degradation  نشان داد  .(adjusted P=0.00015)افزایش بیانTENT4A  با کاهش بقای بیماران همراه بود (Log-rank P<0.05) و توان تشخیصی بالایی داشت (AUC=0.87)،  .(P<0.0001)در حالی کهTENT4B  دارای قدرت تشخیصی متوسط بود (AUC=0.72)، (P=0.01) . همچنین نتایج Real-Time PCR  افزایش معنی‌دار بیان این ژن‌ها را در رده‌های سلولی سرطان کولورکتال نسبت به سلول نرمال نشان داد  .(P<0.05) نتیجه‌گیری: به نظر می رسد کهTENT4A  می‌تواند به عنوان یک بیومارکر بالقوه تشخیصی و پیش‌آگهی‌دهنده در سرطان کولورکتال مطرح باشد و در کنار شواهد موجود، هدف مناسبی برای مطالعات عملکردی و توسعه راهکارهای درمانی هدفمند در آینده محسوب شود.</div> <div style="text-align: justify;">Background and Aim: Colorectal cancer (CRC) is one of the most prevalent malignancies worldwide, and identifying novel molecular biomarkers is essential for early diagnosis and prognostic prediction. TENT4A and TENT4B belong to the family of non- canonical poly (A) polymerases involved in RNA stability and translational regulation. This study aimed to investigate the expression patterns of these genes and their associated signaling pathways in colorectal cancer. Methods: This descriptive- analytical study was conducted using RNA sequencing (RNA-Seq) data obtained from The Cancer Genome Atlas (TCGA). Differentially expressed genes (DEGs) between tumor and normal tissues were identified using the edgeR package. Functional enrichment analysis was subsequently performed using the KEGG and MSigDB databases. The association between gene expression and patient survival was evaluated using Kaplan–Meier survival analysis and Cox proportional hazards regression. Diagnostic performance was assessed using receiver operating characteristic (ROC) curve analysis. In addition, the expression levels of TENT4A and TENT4B were experimentally validated in the HT29 and HCT116 colorectal cancer cell lines relative to HEK293 cells using quantitative real- time PCR (qRT-PCR). Results: A total of 4,558 differentially expressed genes were identified (FDR < 0.05). TENT4A and TENT4B were significantly upregulated, whereas TENT5A, TENT5B, and TENT5C were significantly downregulated (FDR < 0.05). Functional enrichment analysis revealed a significant association of these genes with the RNA degradation pathway (adjusted P = 0.00015). Elevated TENT4A expression was significantly associated with poorer overall survival (log-rank P < 0.05) and demonstrated good diagnostic performance (AUC = 0.87, P < 0.0001), whereas TENT4B exhibited moderate diagnostic accuracy (AUC = 0.72, P = 0.01). Furthermore, qRT-PCR confirmed significantly increased expression of both genes in colorectal cancer cell lines compared with HEK293 cells (P < 0.05). Conclusion: TENT4A appears to be a promising diagnostic and prognostic biomarker for colorectal cancer.
