fa گیرنده‌های کموکاین به‌عنوان اهداف درمانی نوظهور: مروری بر ساختار، عملکرد و کاربردهای بالینی Chemokine receptors as novel therapeutic targets: a review of structure, function, and clinical applications فارماکولوژی Pharmacology مروری Review سابقه و هدف: کموکاین‌ها و گیرنده‌های آن‌ها از تنظیم‌کننده‌های کلیدی مهاجرت سلولی، هدایت لکوسیتی و سازمان‌دهی پاسخ‌های ایمنی محسوب می‌شوند و در طیف وسیعی از فرآیندهای فیزیولوژیک و پاتولوژیک نقش دارند. گیرنده‌های کموکاین عمدتاً به خانواده گیرنده‌های جفت‌شونده با پروتئین G (GPCRs) تعلق داشته و شامل زیرخانواده‌های CCR، CXCR،  CX3CR و XCR هستند. علاوه بر گیرنده‌های تیپیک، گیرنده‌های کموکاین آتیپیک (ACKRs) نیز از طریق تنظیم گرادیان، توزیع و دسترسی کموکاین‌ها در کنترل پاسخ‌های ایمنی و التهابی مشارکت دارند. شواهد فزاینده نشان می‌دهد که محورهای کموکاینی علاوه بر مهاجرت لکوسیتی، در التهاب مزمن، بیماری‌های خودایمنی، عفونت HIV، رگ‌زایی، پیشرفت تومور و متاستاز نقش دارند. روش کار: در این مقاله مروری، مطالعات مرتبط و انتشار یافته تا انتهای سال 2025 ( با تاکید بر مقالات 10 سال اخیر) با استفاده از جست و جوی واژگان کلیدی از جمله “chemokine receptors”، “CXC receptors”، “CCR”، “ACKR”، “chemokine signaling” و “chemokine-targeted therapy در پایگاه‌های اطلاعاتی PubMed و Google Scholar مورد بررسی قرار گرفت. مقالات کاملا مرتبط با اهداف این مقاله مورد بررسی قرار گرفتند. نتیجه گیری: در سال‌های اخیر، مفاهیمی مانند سیگنالینگ سوگیرانه، گیرنده‌های آتیپیک، لیگاندهای آلوستریک و فتوفارماکولوژی، افق‌های جدیدی را در توسعه درمان‌های دقیق‌تر و انتخابی‌تر ایجاد کرده‌اند. در مجموع، درک بهتر زیست‌شناسی و سیگنالینگ گیرنده‌های کموکاین می‌تواند به توسعه نسل جدیدی از درمان‌های هدفمند در بیماری‌های التهابی، بدخیمی‌ها و اختلالات ایمنی منجر شود.   Background and Aim: Chemokines and their receptors are key regulators of cell migration, leukocyte trafficking, and immune response organization, playing critical roles in a wide range of physiological and pathological processes. Chemokine receptors primarily belong to the family of G protein-coupled receptors (GPCRs) and include the CCR, CXCR, CX3CR, and XCR subfamilies. In addition to conventional chemokine receptors, atypical chemokine receptors (ACKRs) regulate chemokine gradients, distribution, and availability, thereby modulating immune and inflammatory responses. Increasing evidence indicates that chemokine signaling pathways are involved not only in leukocyte migration but also in chronic inflammation, autoimmune diseases, HIV infection, angiogenesis, tumor progression, and metastasis. Methods: In this review article, relevant studies published up to the end of 2025 (with an emphasis on articles from the last 10 years) were reviewed using keyword searches including “chemokine receptors”, “CXC receptors”, “CCR”, “ACKR”, “chemokine signaling”, and “chemokine-targeted therapy” in PubMed and Google Scholar databases. Articles that were fully relevant to the objectives of this article were reviewed. Conclusion: Recent advances in biased signaling, atypical chemokine receptors, allosteric ligands, and photopharmacology have opened new perspectives for the development of more selective and precise therapeutic interventions.
