بررسی اثر تیمار با میکروپارتیکل های کیتین بر بار انگلی لیشمانیا ماژور در موشهای Balb/c به روش تهیه رقتهای متوالی

<p>سابقه و هدف: میکرو پارتیکل های کیتین قابلیت تعدیل پاسخهای ایمنی را دارند . به منظور بررسی قابلیت ایمونوادجوانتی کیتین در عفونت با لیشمانیا ماژور ، میزان بار انگلی در موشهای Balb/c آلوده به لیشمانیا ماژور وتحت تیمار با میکروپارتیکل های کیتین موررد ارزیابی قرار گرفت .در اندازه گیری بار انگلی از روش تهیه رقتهای متوالی که دقیق ترین ارزیابی اثربخشی روشهای درمانی و ارزیابی واکسیناسیون در مدل تجربی عفونت لیشمانیا است ، استفاده شد. روش بررسی: موش های Balb/c با 105 × 2 پروماستیگوت انگل لیشمانیا ماژور در فاز ایستایی، در قاعده دم و به طریقه داخل جلدی آلوده شدند و پس از هشت هفته با استفاده از روش تیتراسیون سلولی ، میزان بار انگلی در گروه تحت تیمار ( 6 سر موش)که به صورت دو روز در میان µg/ml 100میکروپارتیکل به صورت زیرجلدی در قاعده دم دریافت کردند و گروه کنترل ( 6 سر موش) که PBS دریافت کردند، تعیین و مقایسه شد . از نطر زمان بروز زخم و وسعت ضایعات هم دو گروه با هم مقایسه شدند. یافته ها: بروز زخم در موشهای تحت تیمار نسبت به موشهای کنترل با تاخیر معنی دار (p<0.02 ) همراه بود و وسعت ضایعات ایجاد شده در موشهای دریافت کننده میکروپارتیکل کوچکتر بود . بار انگلی با وجود اینکه در موشهای تحت تیمار کمتر بود ولی تفاوت آماری معنی دار بین گروه تست وکنترل دیده نشد(P<0.88) . بحث و نتیجه گیری: تلفیق دو روش بررسی ضایعه وسنجش بار انگلی برآورد دقیق تر وصحیحی از وضعیت عفونت و پاسخ ایمنی ضد آن ارائه می دهد. اگرچه حذف انگل راه یافته به احشاء تحت تاثیر تیمار با کیتین دیده نشد اما بروز زخم و وسعت ضایعات تحت تاثیر قرار گرفت. بررسی آثار ایمونوادجوانتی میکروپارتیکل های کیتین در مدلهای عفونت لیشمانیایی نیاز به مطالعات بیشتر دارد.</p>
<p><strong>Background:</strong> Chitin micro-particles have the ability to modulate immune responses. In order to investigate the immunoadjuvant potential of chitin, the parasite burden in Balb/c mice infected with <em>Leishmania Major</em> under treatment with chitin microparticles was evaluated. In measuring the parasitic load, titration method, which is the most accurate assessment of the effectiveness of therapies and vaccine evaluation in experimental models of <em>Leishmania</em> infection was used.</p>
<p><strong> Methods:</strong> Balb/c mice were intradermally infected with 2 × 10<sup>5 </sup>stationary phase of <em>Leishmania Major</em> promastigotes into their base of the tail and eight weeks later, parasite burden were measured and compared between test (6 mice/group) that recived 100 μg/ml microparticle subcutaneously at the base of the tail every two days and control groups (6 mice/group) that received PBS. The onset and size of ulcer among two groups were compared as well.</p>

<p><strong>Results:</strong> Onset of ulcers in the treated mice compared to control showed a significant delay (p<0.02) and the lesion development in the micro-particle recipient mice were significantly smaller. Although the parasite load in treated mice was lower but statistically significant differences were not found between the test group and control (p<0.88).</p>
<p><strong>Conclusion:</strong> The combined use of monitoring the size of the cutaneous lesions and the determination of the parasite burden will reflect more accurately the status of disease. Although the removal of the viseralized parasite was not found among chitin trated group, but onset and size of ulcer was impressed. Immunoadjuvant effects of chitin microparticles in <em>Leishmania</em> infection models requires further studies.</p>
