fa اثر توأم داروی بایولوزاکس- اچ و مکمل زینک برروند اسپرماتوژنز و میزان بیان ژن‌های DAZL و AR در بافت بیضه موش صحرایی Combined Effect of Biolux-H and Zinc Supplementation on Spermatogenesis and the Expression of DAZL and AR Genes in Rat Testicular Tissue زیست شناسی Biology پژوهشی Original <h4 style="text-align: justify;">سابقه و هدف: باروری در مردان شامل توانایی تولید اسپرم  نرمال به تعداد کافی است، عوامل متعددی در روند اسچرماتوژنز و کیفیت پارامترهای اسپرم تاثیرگذار است.  از این عوامل می‌توان به مصرف بعضی داروهای شیمیایی ازجمله زینک و بایولوزاکس– اچ ( ترکیب لوزارتان + هیدروکلروتیازید) از داروهای رایج  در درمان فشار خون اشاره کرد. پژوهش  حاضر به بررسی اثر این داروها براسپرماتوژنز و بیان دوژن مؤثر بر آن در بیضه رت پرداخته است. روش کار: در این مطالعه تجربی32 سر موش نژاد ویستار 10-12 هفته‌ای تهیه و به طور تصادفی به چهار گروه تقسیم شد (8 =n). گروه شاهد دریافت آب مقطر، گروه تیمار بایولوزاکس– اچ (Lnh) با ترکیب (5/12/50 گرم/کیلوگرم وزن بدن از هیدروکلروتیازید ولوزارتان) و گروه تیمار روی (Zn) با (30 میلی‌گرم بر کیلوگرم وزن بدن) وگروه (LnhZn) (20 میلی‌گرم بایولوزاکس- اچ + روی) به صورت توام و به روش (مری- معده‌‌ ای) و به مدت 60 روز، ادامه یافت.پس از اتمام دوره، بافت بیضه، پارامترهای اسپرمی و میزان بیان ژن‌های AR و DAZL در تمامی گروه‌ها بررسی شد. تحلیل واریانس یک طرفه آنوا و آزمون‌های توکی انجام شد.   یافته‌ها: در گروه Lnh کاهش معنادار در تعداد  سلول‌های اسپرماتید(8/1 ± 34/1) در مقایسه با کنترل مشاهده شد. در گروه Zn، همه عوامل مشابه گروه کنترل بودند و تعداد اسپرماتوسیت ها (1/3 ± 75/11) و سلول‌های سرتولی (3/4 ± 14/63) در این گروه‌هادر مقایسه با کنترل افزایش یافت اما این افزایش تنها در تعداد اسپرماتوسیت‌ها به صورت معنادار مشاهده شد. (0/05p<) گروه LnhZn نتایج مشابهی را در تمام عوامل در مقایسه با گروه کنترل داشت، همچنین بیان ژن‌های  AR(0/001± 0/0048) و DAZL (0/006 ± 0/008) درگروه Lnh نسبت به کنترل (0/04 ± 1)کاهش معنادار داشت (0/05= p). ژن AR در گروه Zn (0/073 ± 1/247) و LnhZn (0/224± 1/354) در مقایسه با کنترل افزایش بیان را نشان داد و ژن DAZL نیز در گروه‌های تجربی Zn با مقدار (0/4 ± 1/68) و گروه LnhZn (0/54 ± 1/58) افزایش را به صورت معنادار نشان داد (0/05p<). نتیجه‌گیری: به نظر می‌رسد استفاده از مکمل روی سبب بهبود عوارض ناشی از مصرف  بایولوزاکس- اچ بر کیفیت اسپرم شد.  </h4> <div style="text-align: justify;">Background and Aim: Fertility in men includes the ability to produce a sufficient number of normal sperm. Several factors affect the schermatogenesis-quality of sperm parameters. These factors include the use of certain chemical drugs, such as zinc and bioluxax-H (a combination of losartan + hydrochlorothiazide), common drugs used to treat hypertension. The present study was carried out to investigate the effect of these drugs on spermatogenesis and the expression of Dogen on its effect in rat testis. Methods: In the present experimental study, 32 Wistar rats were randomly divided into four groups (n= 8): Group I (control) received distilled water, Group II (Lnh) was treated with Lnh (losartan and hydrochlorothiazide) (20/5 mg/kg BW/day, Group III (Zn) was treated with the Zn (Zinc gluconate, 30mg/kg BW/day, and Group IV (LnhZn) was treated with Lnh and Zn at the same doses. Treatment was continued in all groups for 60 days. After the treatment, testicular tissue, sperm parameters, and AR and Dazl genes expression in the rats were investigated. Results: In Lnh group, a significant decrease in spermatid cell count (34.1 ± 1.8) was observed as compared to the controls. In Zn group, all factors were similar to those in the control group and spermatocyte count (75.11 ± 1.3) and Sertoli cells (14.62 ± 3.4) increased in these groups compared to those of the controls; however, this increase was observed only in the number of spermatocytes (p < 0.05). Also, the LnhZn group had similar results in all factors compared to the control group. Moreover, the expression of AR (0.0048 ± 0.001) and DAZL (0.008 ± 0.006) genes in the Lnh region was significantly reduced compared to that in the control group (1 ± 0.04) (p= 0/05). AR gene showed increased expression in Zn (1.247 ± 0.073) and LnhZn (1.354 ± 0.224) groups compared to the controls. In addition, DAZL gene in Zn experimental groups (1.68 ± 0.4) and LnhZn (1.58 ± 0.54) showed a significant increase (p < 0/05).
