fa بررسی اثر نانوامولسیون روغن کلزا بر فاکتورهای هماتولوژیک موش مبتلا به سرطان پستان با رده سلولی TUBO Investigating the Effects of Canola oil Nanoemulsion on Hematological Parameters of Mice with Breast Cancer Cell Line TUBO فیزیولوژی Physilogy پژوهشی Original <div style="text-align: justify;">سابقه و هدف: در کشورهای در حال توسعه شیوع سرطان پستان روند افزایشی داشته و در حال تبدیل شدن به یک مشکل بزرگ جهانی است. تحقیقات گسترده‌ای به درک علل سرطان پستان و ایجاد روش‌های درمانی جدید برای این بیماری اختصاص یافته است.  گیاه کلزا غنی از ترکیبات حاوی گوگرد است که مسئول محتوای غنی از گلوکوزینولات، بو و عطر و طعم تند است. این گیاه حاوی آنتی‌اکسیدان‌هایی مانند ویتامین C و E، کاروتنوئیدها و آنزیم‌های آنتی‌اکسیدان مانند کاتالاز، سوپراکسید دیسموتاز (SOD) و پراکسیداز است. هدف از این تحقیق بررسی اثر نانوامولسیون روغن کلزا بر فاکتورهای هماتولوژیک موش مبتلا به سرطان پستان با رده سلولی TUBO است. روش کار: در این تحقیق تجربی سنتز نانوامولوسیون از روغن کلزا انجام شد و تایید اندازه و پایداری آن انجام  شد. سپس سلول‌های سرطانی TUBO کشت داده شده و به موش‌ها تزریق شدند. تیمار با نانوامولوسیون روغن کلزا با غلظت‌های 1، 2 و 4 درصد و همجنین با تاموکسیفن انجام  شد. تجزیه و تحلیل آماری داده‌های  مرتبط با پارامترهای خونی به وسیله نرم‌افزار آماری SPSS انجام و برای مقایسه نتایج بین گروه‌ها از آنالیز واریانس یک طرفه و تست توکی استفاده شد. 05/0 p < معیار معناداری در نظر گرفته شد. یافته‌ها: در تحقیق حاضر میزان گلبول‌های سفید در گروه سرطانی نسبت به گروه کنترل افزایش معناداری داشته است (کنترل: 93 ± 3167 سرطانی: 676 ± 8150 و 0/01P < ). با انجام تیمار و افزایش میزان نانوامولوسیون روغن کلزا میزان گلبول‌های سفید نسبت به گروه سرطانی کاهش یافته است (1557 ± 4775 و 0/01P < ). برعکس این اتفاق برای گلبول‌های قرمز شکل گرفته است و از 400000 ± 7712000 در گروه کنترل به 190000 ± 5908000 در گروه سرطانی کاهش یافته است (0/001P < ). تیمار با نانوامولسیون سبب اصلاح این تغییر شده و تعداد گلبول‌های قرمز ر ا به 120000 ± 6444000 رسانده است (0/05P < ). در مورد هموگلوبین و پارامترهای مرتبط با آن اختلاف معناداری بین گروه‌های کنترل و سرطانی و تحت درمان با نانوامولسیون و تاموکسیفن مشاهده نشد. تعداد پلاکت‌ها در گروه سرطانی تفاوت معناداری با گروه کنترل نشان داد و گروه‌های تحت درمان دچار تغییرات معنادار در جهت نزدیک شدن به گروه کنترل نشان دادند (کنترل: 131243 ± 787000، سرطانی: 40038 ± 405250 و تحت درمان با نانوامولسیون: 48264 ± 633400 (05/0P < ). نتیجه‌گیری: نانوامولسیون روغن کلزای سنتز شده با سایز زیر 50 نانومتر و پتانسیل زتای حدود 30- دارای پایداری بالا و پراکندگی یکنواخت است. القای سرطان سبب تغییراتی در پارامترهای خونی موش‌ها شد. تیمار با نانوامولسیون درحد مطلوبی این تغییرات را اصلاح کرد که احتمالآ این قابلیت مرتبط با ترکیبات آنتی‌اکسیدانی موجود در روغن کلزا باشد.</div> <div style="text-align: justify;">Background and Aim: In developing countries, the prevalence of breast cancer has increased and is becoming a major global problem. Extensive research has been devoted to understanding the causes of breast cancer and creating new treatment methods for this disease. The canola plant is rich in sulfur-containing compounds responsible for its rich content of glucosinolates, pungent aroma, and flavor. This plant contains antioxidants such as vitamin C and E, carotenoids, and antioxidant enzymes such as catalase, superoxide dismutase (SOD), and peroxidase. This research aims to investigate the effect of canola oil nanoemulsion on the hematological factors of mice suffering from breast cancer with the TUBO cell line. Methods: In this experimental research, the synthesis of nanoemulsion from canola oil was carried out, and its size and stability were confirmed. Then the TUBO cancer cells were cultured and injected into mice. The treatment was carried out with canola oil nanoemulsion with 1, 2 and 4% concentrations and homogenous with tamoxifen. Statistical analysis of data related to blood parameters was done by SPSS statistical software .One-way ANOVA and Tukey's test were used to compare the results between groups (p<0.05 was considered significant). Results: In the present study, the count of white blood cells in the cancer group increased significantly compared to the control group (control: 3167 ± 93, cancer: 8150 ± 676, P < 0.01). Furthermore, by performing the treatment and increasing the amount of Canola oil nanoemulsion, the amount of white blood cells decreased compared to the cancer group (4775 ± 1557 and P < 0.01). On the contrary, the count of red blood cells decreased from 7712000 ± 400000 in the control group to 5908000 ± 190000 in the cancer group (P < 0.001). Treatment with nanoemulsion corrected this change and increased the number of red blood cells to 6444000 ± 120000 (P < 0.05). Regarding hemoglobin and related parameters, no significant difference was observed between the control and the cancer groups and treatmentwith nanoemulsion and tamoxifen groups. The number of platelets in the cancer group showed a significant difference from the control group, and the treated groups showed significant changes = somewhat similar to the control group (control: 787000 ± 131243, cancer: 405250 ± 40034, treated with nanoemulsion: 633400 ± 48264 (P < 0.05). Conclusion: The synthesized Canola oil nanoemulsion with the size below 50 nm and a zeta potential of about -30 has high stability and uniform dispersion. Cancer induction caused changes in the blood parameters of mice.
