fa بررسی تأثیر آنتی‌ژن توموری MAGE-A3 برفعالیت اونکولایتیکی ویروس رئو در رده‌های سلول توموری کولورکتال Evaluation of the Effect of MAGE-A3 Tumor Antigen on the Oncolytic Activity of Reovirus in Colorectal Cancer Cell Lines ویروس شناسی Virology پژوهشی Original <div style="text-align: justify;">سابقه و هدف: سرطان کولورکتال سومین دلیل بدخیمی شایع و چهارمین دلیل مرگ‌ومیر ناشی از سرطان در سراسر جهان است .اگرچه بسیاری از درمان‌ها از جمله جراحی، پرتودرمانی و شیمی‌درمانی برای آن استفاده می‌شود، ولی با توجه به مؤثر نبودن آنها تحقیق‌ها روی سایر روش‌ها درحال انجام است. استفاده از آنتی‌ژن‌های توموری علیه تومور یکی از انواع روش‌های درمانی است که در همراهی با سایر روش‌های درمانی می‌تواند سبب تحریک بهتر سیستم ایمنی علیه تومور شود. استفاده از ویروس‌های انکولایتیک با ویژگی طبیعی در از بین بردن سلول‌های سرطانی زمینه گسترده‌ای را برای مطالعه‌های بیشتر فراهم کرده است. هدف از این پژوهش بررسی تأثیر آنتی‌ژن توموری MAGE-A3 بر فعالیت اونکولایتیکی ویروس رئو در رده‌های سلول توموری کولورکتال است. روش کار: در این مطالعه تجربی از سلول L929 که جزو رده سلولی فیبروبلاست است به عنوان سلول میزبان برای تکثیر رئوویروس استفاده شد. تیتر رئو ویروس با روش CCID50 تعیین و تیتر مناسب برای مطالعه انتخاب شد. پلاسمید بیان کننده MAGEA3 به سلول‌های سرطان کولورکتال با منشأ موشی (CT26) و انسانی (Caco2) ترانسفکت شده و با ویروس رئو (1 = MOI و 01/0 = MOI) آلوده شدند. 24 ساعت بعد از تلقیح رئوویروس و مشاهده CPE سلول‌ها برای بررسی آپوپتوز جمع شدند و با روش فلوسایتومتری سنجیده شدند. یافته‌ها: میزان آپوپتوز در سلول CT26 در حضور وکتور بیان‌کننده MAGE-A3 و آلوده شده با ویروس رئو (در 1 = MOI 3/8 ± 38/5درصد و  0/02 > p-value / در 0/01 = MOI 2/1 ± 30 درصد و 0/005 > p-value) نسبت به نبود وکتور بیان‌کننده MAGE-A3، (در 1 = MOI 0/9 ± 22/4درصد و 0/01 = MOI 0/3 ± 15/8درصد) بیشتر بود. از طرفی میزان آپوپتوز در سلول Caco2 در حضور وکتور بیان‌کننده MAGE و آلوده شده با ویروس رئو (در 1 = MOI 1/8 ± 22/5درصد و/ p-value = NS  در 0/01 = MOI 2 ± 11 درصد و p-value = NS) نسبت به نبود وکتور بیان‌کننده MAGE-A3، (در 1 = MOI 2/7 ± 45/5 درصد و 0/01 = MOI 0/9 ± 11/5درصد) کمتر بود. می‌توان چنین بیان کرد که با توجه به نتایج حاصل از این مطالعه تأثیر سیتولیز توسط  ویروس رئو در رده سلولیCaco2 بیشتر از سلول CT26 است و همچنین میزان کشندگی سلول در حضور وکتور بیان‌کننده MAGE-A3 در سلول CT26 تقریبا دو برابر حالتی است که این وکتور در سلول Caco2 وجود دارد. نتیجه‌گیری: با توجه به میزان کشندگی بالاتر ویروس در سلول‌های Caco2 و وضعیت ژن‌های P53 و Ras در این سلول به نظر می‌رسد که P53 موتانت ولی Ras فعال است، ویروس فعالیت بیشتری داشته و حضور  MAGE-A3با کاهش فعالیت Ras، از فعالیت ویروس می‌کاهد.  </div> Background and Aim: Colorectal cancer (CRC) is the third most deadly and fourth most commonly diagnosed cancer in the world. Although many interventions such as surgery, radiation therapy and chemotherapy are used nowadays, researchon other methods is being carried out too. The use of tumor antigens against the tumor is a kind of treatment that in combination with the virus can better stimulate the immune system against the tumor. The use of oncolytic viruses with natural characteristics in destroying cancer cells has provided a wide field for further studies. The aim of this study was to investigate the effect of MAGE-A3 tumor antigen on the oncolytic activity of Reovirus in colorectal cancer cell lines. Methods: In this experimental study L929, a type of mouse fibroblast cell line, was used as a host cell for Reovirus propagation. Reovirus titer was determined by CCID50 method and the appropriate titer was selected for the study. The MAGEA3- expressing plasmid was transfected into colorectal cancer cells of the mouse (CT26) and human (Caco2) origin and infected with Reovirus (MOI = 1 and MOI = 0.01). 24 hours after Reovirus inoculation and observing CPE, the cells were collected to investigate apoptosis and measured by flow cytometry. Results: The apoptosis rate in CT26 cells in the presence of MAGE-A3 expressing vector and inoculated with Reovirus was higher (MOI=1, 38.5 ± 3.8% / p-value < 0.02 and in MOI=0.01, 30±  2.1% / p-value < 0.005) in comparison to the absence of MAGE-A3 expressing vector (MOI=1, 22.4±  0.9 and MOI=0.01, 15.8 ± 0.3). On the other hand, the rate of apoptosis in Caco2 cells in the presence of MAGE-A3 expressing vector and inoculated with Reovirus (MOI=1, 22.5±  1.8% / p-value=ns and in MOI=0.01, 11 ± 2% / p-value=ns) was lower than in the absence of vector expressing MAGE-A3 (MOI=1, 45.5±  2.7 and MOI=0.01, 11.5±  0.9). It can be stated that according to the results of this study, the cytolysis effect of Reovirus in Caco2 cell line is higher than that of CT26 cell and also the rate of cell cytolysis in the presence of MAGE expressing vector is almost twice more than that of this vector in Caco2 cells.
