fa تأثیر عصاره هیدروالکلی صمغ ریشه و ساقه گیاه آنغوزه Ferula assa- foetida بر بیان ژن‌های VEGFو FGF در پرده کوریوآلانتوئیک جنین جوجه The Effect of the Hydroalcoholic Extract of Ferula Assa- Foetida Resin Root and Shoot on the VEGF and FGF Gene Expression in the Chorioallantoic Membrane of Chick Embryos زیست شناسی Biology پژوهشی Original سابقه و هدف: آنژیوژنز فرایند زیستی جوانه زدن رگ‌های جدید از رگ‌های موجود در بافت است. عامل اصلی در هدایت مولکولی این فرایند، فاکتورهای رشد اندوتلیال عروقی (VEGF) و فاکتور رشد فیبروبلاستی (FGF) هستند. با توجه به اهمیت علوم گیاه پزشکی و از طرفی نقش رگزایی در فرآیند‌هایی مانند ترمیم زخم، سیکل‌های قاعدگی، رشد جفت و تخمک‌گذاری، هدف این پژوهش بررسی اثر عصاره هیدروالکلی ریشه و ساقه گیاه آنغوزه (به عنوان یکی از گیاهان دارویی شاخص در طب سنتی) بر تغییرهای بیان ژن‌های VEGF و FGF در مسیر آنژیوژنز پرده کوریوآلانتوئیک (CAM) جنین جوجه بود. روش کار: در این پژوهش تجربی، تعداد 80 عدد تخم‌مرغ نطفه‌دار نژاد Ross به طور تصادفی به هشت گروه شامل گروه شاهد، شاهد آزمایشگاهی (pbs)، و شش گروه تجربی تقسیم شدند. در روز هشتم انکوباسیون، گروه شاهد آزمایشگاهی با نرمال سالین و گروه‌های تجربی با دوزهای ۱۰۰، ۲۰۰ و ۳۰۰ میکروگرم بر میلی‌لیتر عصاره هیدروالکلی ساقه و ریشه گیاه آنغوزه تیمار شدند. در روز دوازدهم، عکس‌برداری از پرده کوریوآلانتوئیک برای شمارش تعداد عروق و اندازه‌گیری طول عروق انجام شد. تهیه نمونه CAM برای استخراج RNA و ساخت cDNA انجام گرفت. داده‌های جمع‌آوری شده توسط آزمون‌های آماری t- test و ANOVA بررسی شدند. یافته‌ها: میانگین تعداد عروق ( 45/3 ± 1/ 26) و طول عروق (0/78± 31/1) در گروه‌ تیمارشده کاهش معناداری) 0/05< P) را نسبت به گروه شاهد نشان داد. همچنین میزان بیان ژن‌های VEGF (0/3045) و FGF (0/4229) در گروه تیمارشده به طور معناداری ) 0/05< P) کمتر از گروه شاهد بود. نتیجه‌گیری: به‌نظر می‌رسد عصاره هیدروالکلی گیاه آنغوزه می‌تواند بر میزان بیان ژن‌های  VEGFو FGF و روند رگ‌زایی پـرده کوریوآلانتوئیک جنین‌ جوجه تأثیر گذارد. Background and Aim: Angiogenesis is the biological process of sprouting new vessels from existing vessels in the tissue. The main factor in the molecular guidance of this process is vascular endothelial growth factors (VEGF) and fibroblast growth factor (FGF). Considering the importance of plant medicine sciences and also, the role of angiogenesis in the processes such as wound healing, menstrual cycles, placental growth, and ovulation, the aim of this study was to investigate the effect of hydroalcoholic extract of the root and shoot of Ferula assa- foetida plant (as one of the prominent medicinal plants in traditional medicine) on changes in the gene  expression of VEGF and FGF in the angiogenesis pathway of the chorioallantoic membrane (CAM) of chick embryos. Methods: In this experimental research, 80 Ross fertilized eggs of the were randomly divided into 8 groups including the control group, laboratory control (pbs), and 6 experimental groups. On the eighth day of incubation, the laboratory control group was treated with normal saline and the experimental groups were treated with doses of 100, 200 and 300 µg/ml of the hydroalcoholic extract of the shoot and root of Ferula assa- foetida plant. On the twelfth day, a photo was taken of the chorioallantoic membrane to count the number of vessels and measure the length of the vessels. CAM samples were prepared for RNA extraction and cDNA production. The collected data were analyzed by t-test and ANOVA. Results: The average number of vessels (26.1 ± 3.45) and vessel length (31.1 ± 0.78) in the treated group showed a significant decrease (P < 0.05) compared to the control group. Also, the gene expression level of VEGF and FGF in the treated group was significantly (P < 0.05) lower than the control group.
