fa بررسی ارتباط پلی‌مورفیسم (C29T (Pro10Leu ژن Transforming Growth Factor βeta1 (TGF-β1) و ناباروری با علت نامشخص در ایران ایمنی‌شناسی Immunology پژوهشی Original <div style="text-align: right"> <p dir="RTL">چکیده</p> <p dir="RTL">سابقه و هدف: ناباروری ازجمله بیماری‌هایی محسوب می‌شود که شیوع روزافزون و نگران‌کننده‌ای دارد. ده الی سی درصد از ناباروری‌ها به‌عنوان با علت نامشخص (<span dir="LTR">UI<a href="#_ftn1" name="_ftnref1" title="">[1]</a></span>) دسته‌بندی می‌شوند. <span dir="LTR">UI</span> یک وضعیت مطلق ازلحاظ بالینی نیست بلکه به ناتوانی در باردارشدن یا زنده‌زایی گفته می‌شود. <span dir="LTR">TGF-&beta1</span> سایتوکاینی با عملکرد تنظیم ایمنی قوی است و فرایند لانه گزینی و اتصال سلول‌های تروفوبلاست به ماتریکس خارج سلولی را تنظیم می‌کند. در این مطالعه ارتباط پلی‌مورفیسم (<span dir="LTR">C29T (Pro10Leu</span> ژن <em><span dir="LTR">TGF-&beta1</span></em> و ناباروری با علت نامشخص در بیماران ایرانی موردبررسی قرار گرفت.</p> <p dir="RTL">مواد و روش‌ها: در این مطالعه مورد-شاهد نمونه‌های خون از ۱۴۲ مرد و ۳۵ زن نابارور و ۲۳۲ مرد و ۱۰۴ زن سالم و بدون سابقه ناباروری (گروه کنترل) جمع‌آوری شدند<span dir="LTR">.</span> استخراج <span dir="LTR">DNA</span> توسط روش حذف نمک<a href="#_ftn2" name="_ftnref2" title=""><span dir="LTR">[2]</span></a> و تکثیر قطعه حاوی پلی‌مورفیسم موردنظر توسط روش <span dir="LTR">PCR<a href="#_ftn3" name="_ftnref3" title="">[3]</a></span> انجام شد. برای بررسی پلی‌مورفیسم<span dir="LTR">C29T (Pro10leu)</span> ژن <em><span dir="LTR">TGF-&beta1 </span></em>&nbsp;و تعیین ژنوتیپ افراد از روش <span dir="LTR">Automated Sequencing</span> استفاده گردید.</p> <p dir="RTL">یافته‌ها: اختلاف معناداری در فراوانی توزیع ژنوتیپ‌ها و آلل‌های مختلف پلی‌مورفیسم (<span dir="LTR">C29T (Pro10Leu</span> در مقایسه بین گروه‌های مختلف بیماران و گروه کنترل و همچنین بررسی زوج‌های نابارور و کنترل مشاهده نشد (<span dir="LTR">P value</span> > ۰٫۰۵).</p> <p dir="RTL">نتیجه‌گیری: نتایج این مطالعه نشان می‌دهد بین ناباروری با علت نامشخص و پلی‌مورفیسم (<span dir="LTR">C29T (Pro10Leu</span> ژن <em><span dir="LTR">TGF-&beta1</span></em> ارتباط معناداری وجود ندارد. برای مشخص شدن رابطه‌ی این پلی‌مورفیسم و استعداد ابتلا به ناباروری با علت نامشخص، به مطالعات مرتبط بیشتری نیازمندیم.</p> <p dir="RTL"></p><p dir="RTL"></p> <div>&nbsp; <div id="ftn3"></div></div> </div> <p class="MsoNormal" style="line-height: 115%"><font face="times new roman, times, serif"><b><span style="font-size: 12pt line-height: 115%">Abstract:</span></b><b><span style="font-size: 12pt line-height: 115%"></span></b></font></p> <p class="MsoNormal" style="line-height: 115%"></p> <p class="MsoNormal" style="line-height: 115%"><font face="times new roman, times, serif"><b><span style="font-size: 12pt line-height: 115%">Background and Aim</span></b><b><span style="font-size: 12pt line-height: 115%">:</span></b><span style="font-size: 12pt line-height: 115%"> The increasing incidence of infertility is alarming. About 10-30% of infertility are classified as Unexplained Infertility (UI) which is not an absolute clinical condition. </span><span style="font-size: 12pt line-height: 115%">TGF-</span><span style="font-size: 12pt line-height: 115%">b</span>_{<span style="font-size: 12pt line-height: 115%">1 </span>}<span style="font-size: 12pt line-height: 115%">is multifunctional cytokine and produced mainly by T regulatory (Treg) lymphocytes. This cytokine plays important roles in physiology of normal pregnancy. The association of Single Nucleotide <span style="text-transform: uppercase">P</span>olymorphisms (SNPs) of <i>TGF-</i></span><i><span style="font-size: 12pt line-height: 115%">b</span></i><i>_{<span style="font-size: 12pt line-height: 115%">1</span>}</i>_{<span style="font-size: 12pt line-height: 115%">&nbsp;</span>}<span style="font-size: 12pt line-height: 115%">gene with many immunologic diseases has been reported. </span><span style="font-size: 12pt line-height: 115%">In this study, the association of TGF-&beta1 </span><span style="font-size: 12pt line-height: 115%">C29T (Pro10Leu</span><span style="font-size: 12pt line-height: 115%">) gene polymorphism with unexplained infertility was investigated in Iranian UI patients.</span></font></p> <p class="MsoNormal" style="line-height: 115%"><font face="times new roman, times, serif"><b><span style="font-size: 12pt line-height: 115%">Materials and Methods</span></b><span style="font-size: 12pt line-height: 115%">: In this case-control study blood samples were collected from177 UI patients (142 male and 35 female) &nbsp;and 336 controls (232 male and 104 female) with no history of infertility. DNA was extracted by the salting out method.
