fa بیان ژن HER4 درنمونه های بلوک پارافینه بیماران مبتلا به سرطان پستان پژوهشی Original <p dir="RTL"><strong>مقدمه</strong>: سرطان پستان دومین عامل مرگ و میر در دنیا است. درک آسیب شناسی مولکولی سرطان پستان می تواند اطلاعات مفیدی در مورد مسیرهای جدید درمانی در اختیار قرار دهد. ژن <em><span dir="LTR">HER4</span></em> &nbsp;به عنوان نشانگر پیش آگهی مولکولی در سرطان ها مورد توجه قرار گرفته است. بنابراین هدف از این مطالعه بررسی میزان بیان این ژن در افراد مبتلا به سرطان پستان می باشد.</p> <p dir="RTL"><strong>روش ها</strong>: در این مطالعه 70 نمونه بلوک پارافینه شامل 35 نمونه بیمار سرطان پستان و 35 نمونه نرمال جمع آوری گردید. پس از برش گیری و پارافین زدایی،<span dir="LTR">RNA </span>&nbsp;نمونه ها استخراج، سپس سنتز <span dir="LTR">cDNA</span> &nbsp;به کمک آنزیم <span dir="LTR">M-MuLV</span> و پرایمرهای <span dir="LTR">Oligo dt</span> و<span dir="LTR">Random Hexamer </span>&nbsp;انجام شد. آنالیز آماری مطالعه توسط نرم افزار <span dir="LTR">GraphPad prism</span> با روش <span dir="LTR">t-test</span> انجام شد.</p> <p dir="RTL"><strong>یافته ها</strong>: نتایج واکنش <span dir="LTR">Real Time PCR</span> بیانگر، کاهش بیان ژن <em><span dir="LTR">HER4</span></em> &nbsp;در اکثر نمونه ها نسبت به نمونه های نرمال بود و مشخص شد که بیان ژن با پیشرفت مرحله بیماری دارای رابطه معکوس می باشد، به طوری که در نمونه هایی با مرحله بیماری 1 و 2 میانگین بیان ژن <em><span dir="LTR">HER4</span></em> به میزان 11/1&nbsp;&nbsp;&nbsp; &nbsp;<span dir="LTR">(Pvalue= 0.048)</span> نسبت به نمونه نرمال افزایش و در مراحل 3و 4 میانگین بیان ژن به میزان 624/0<span dir="LTR">(Pvalue= 0.048) </span>&nbsp;کاهش یافته است.</p> <p dir="RTL"><strong>نتیجه گیری</strong>: با توجه به نتایج می توان بیان نمود بررسی میزان بیان ژن <em><span dir="LTR">HER4</span></em> &nbsp;می تواند در تشخیص سرطان پستان، به عنوان یک مارکر نقش داشته باشد، همچنین در شناسایی و تشخیص مراحل اولیه بیماری موثر باشد.</p> <p><strong>Background:</strong> the breast cancer is the second cause of worldwide death. Understanding of molecular pathology of breast cancer can provide useful information about new treatment routes. <em>HER4</em> gene considered as a molecular pre-prognostic marker in cancers recently. So the purpose of this research is studying of <em>HER4 </em>gene expression in breast cancer patients.</p> <p><strong>Methods:</strong> in this study 70 samples of paraffin blocks of 35 samples from breast cancer patients and 35 normal samples were collected. After slicing and deparaffinization, RNA of samples were extracted and then cDNA synthesis using of M-MuLV enzyme and Oligo dt and Random hexamer primers were done. Statistical analysis of research by Graph Pad prism software with t-test procedure were done.</p> <p><strong>Findings:</strong> results of Real-time PCR show <em>HER4</em> gene expression decreased in most samples compared to normal ones and It was determined that gene expression has the inverse relation with progression in cancer stage. Therefore, in the samples with first and second stage <em>HER4</em> gene expression increased 1.11(Pvalue0.048) compared with normal ones and in the third and fourth stage <em>HER4</em> gene expression 0.624 (Pvalue0.048) decreased<span dir="RTL">.</span></p> <p><strong>conclusion:</strong> According to the results, the study of <em>HER4</em> gene expression may have a role as a diagnostic marker in breast cancer.
