fa بررسی اثر عصاره اتانولی پونه کوهی بر بیان واسطه‌های پیش التهابی در سلول‌های تک هسته‌ای خون محیطی The Effect of Ethanolic Extract of Mentha Pulegium on the Expression of Pro- Inflammatory Mediators in Peripheral Blood Monocytes فیزیولوژی Physilogy پژوهشی Original <div style="text-align: justify;">سابقه و هدف: التهاب پاسخ اولیه سیستم ایمنی بدن به محرک‌های آسیب‌رسان است. هدف از انجام این مطالعه بررسی اثر عصاره اتانولی پونه کوهی بر بیان ژن واسطه‌های پیش‌التهابی iNOS و NF- kB در سلول‌های تک هسته‌ای جدا شده از خون محیطی انسان فعال شده با لیپوپلی ساکارید است. روش کار: این مطالعه آزمایشگاهی بر روی نمونه خون 12 فرد سالم انجام شد. در گروه اول: بافر PBS به محیط‌های کشت اضافه شد. گروه دوم: لیپو پلی ساکارید برای تحریک التهاب با غلظت ng / ml 10 استفاده شد. گروه سوم: پس از تحریک با لیپوپلی ساکارید، غلظت μg / ml 10 عصاره اتانولی پونه کوهی در محیط کشت استفاده شد. گروه چهارم: پس از تحریک با لیپوپلی ساکارید، غلظت μg / ml 30 عصاره اتانولی پونه کوهی استفاده شد. گروه پنجم: پس از تحریک با لیپو پلی ساکارید، غلظت μg / ml 60 عصاره اتانولی پونه کوهی استفاده شد. در پایان میزان بیان ژن واسطه‌های پیش‌التهابی iNOS و NF- KB در گروه‌های مختلف تعیین شد. خواهد شد. تحلیل داده‌ها با استفاده از نرم‌افزار SPSS آنالیز one way ANOVA و تست تکمیلی توکی انجام شد. یافته‌ها: نتایج داده‌های مطالعه حاضر نشان داد که بیان ژن NF- kB در گروه دوم (0/14 ± 1/09) در مقایسه با گروه اول به طور معناداری افزایش نشان داد (0/05>P). بیان ژن NF- kB در گروه سوم (0/09 ± 0/48)، گروه چهارم (0/13 ± 0/54) و گروه پنجم (0/24 ± 0/66) در مقایسه با گروه دوم به طور معناداری کمتر بود (0/05 >P). در این مطالعه  بیان ژن iNOS در گروه دوم (0/22 ± 1/14) در مقایسه با گروه اول به طور معناداری افزایش نشان داد   (0/05>P). بیان ژن iNOS در گروه سوم (0/24 ± 0/35) و گروه چهارم (0/07 ± 0/29) در مقایسه با گروه دوم اختلاف معناداری نداشت (0/05>P). نتیجه‌گیری: به نظر می رسد که عصاره اتانولی پونه کوهی دارای اثرات ضد التهابی است و می‌تواند در تغییر بیان ژن واسطه‌های پیش‌التهابی iNOS و NF- kB در سلول‌های تک هسته‌ای جدا شده از خون محیطی انسان فعال شده با لیپوپلی ساکارید موثر باشد.</div> <div style="text-align: justify;">Background and Aim: Inflammation is the immune system’s primary response to harmful stimuli. The aim of this study was to evaluate the effect of ethanolic extract of mentha pulegium on the expression of pro- inflammatory mediators of iNOS and NF-κB in mononuclear cells isolated from human peripheral blood activated with lipopolysaccharide. Methods: In this experimental study, mononuclear cells of 12 individuals with no history of specific disease were used. In the first group: PBS buffer was added to the culture media. Group 2: Lipopolysaccharide was used to stimulate inflammation at a concentration of 10 ng/ ml (19). Group 3: After stimulation with lipopolysaccharide, a concentration of 10 μg / ml ethanolic extract of mentha pulegium was used in the culture medium. Group 4: After stimulation with lipopolysaccharide, a concentration of 30 μg / ml ethanolic extract of mentha pulegium was used. Group 5: After stimulation with lipopolysaccharide, a concentration of 60 μg / ml ethanolic extract of mentha pulegium was used. Then, the expression of iNOS and NF-KB pro- inflammatory mediators in different groups wasmeasured. Data analysis was performed using SPSS software with one-way ANOVA analysis and Tukey supplementary test. Results: The results of the present study showedthat the expression of NF-κB gene in the second group (1.09 ± 0.14) significantly increased compared to the first group (P˂0.05). Moreover, NF-κB gene expression was significantly lower in the third group (0.48 ± 0.09), the fourth group (0.54 ± 0.13), and the fifth group (0.66 ± 0.24) compared to the second group (P˂0.05).In addition,, iNOS gene expression in the second group (1.14 ± 0.22) showed a significant increase compared to the first group (P˂0.05). The expression of iNOS gene in the third group (0.35 ± 0.24) and the fourth group (0.29 ± 0.07) was not significantly different from the second group (P˂0.05).
