fa بررسی نقش ژن‌های ‌HLA-A, -B, -DRB1‎‌ با بیماری پریودونتیت Investigating the Role of HLA-A, -B, -DRB1 Genes in Periodontitis Disease ژنتیک Genetic پژوهشی Original <div style="text-align: justify;">سابقه و هدف: پریودونتیت یکی از بیماری‌های التهابی پیرامون دندانی است و شایع‌ترین دلیل از دست دادن دندان محسوب می‌شود. اگرچه عفونت‌های باکتریایی نقش عمده‌ای در پاتوبیولوژی پریودونتیت ایفا می‌کنند، ولی شدت این بیماری‌ها به واسطه شدت و حدت پاسخ ایمنی میزبان به این عفونت‌ها تعیین می‌شود. آنتی‌ژن‌های لکوسیت انسانی (HLAs) در فرایندهای بیولوژیکی گوناگون، از جمله سازگاری بافتی، التهاب، تنظیم پاسخ ایمنی و آبشار کمپلمان نقش ایفا می‌کنند. ما در مطالعه پیش رو به بررسی تفاوت فراوانی آلل‌های لکوس HLA در بیماران مبتلا به پریودونتیت در مقایسه با افراد سالم پرداختیم. روش کار: این مطالعه یک مطالعه مورد- شاهدی است. نمونه‌های بافتی از 40 بیمار مبتلا به پریودنتیت و 40 فرد شاهد سالم گرفته شد. نمونه‌های مورد و شاهد بر اساس معیارهای ورود و خروج مطالعه انتخاب شدند. در نمونه‌های گرفته شده، ژنوتیپ لکوس های HLA-A، HLA-B و HLA-DRB1 با روش SSP و با استفاده از کیت شرکت Olerup تعیین شد. فرکانس آللی به روش شمارش مستقیم تعیین شد و برای آنالیز آماری بین دو گروه از مدل multinomial regression استفاده شد. بسته نرم‌افزاری &rlm;Arlequin&rlm; نسخه 3.5.2.2 &rlm;Excoffier and Lischer, 2010)&lrm;&rlm;&rlm;( و نرم‌افزار R نسخه 1/6/3 برای انجام محاسبات آماری استفاده شد. <a name="_Hlk127109546">آزمون آماری کای دو و </a>OR نیز محاسبه شد. یافته‌ها: این مطالعه روی 40 بیمار و 40 شاهد انجام شد. افراد دو گروه مشابه بودند. فراوانی آلل DRB1*04 در گروه مورد (28 مورد) به شکل معناداری بیش از گروه شاهد (16 کنترل) بود (0/01 > P) Odd Ratio محاسبه شده برای این آلل برابر 2/3 بود (2/3 = OR). فراوانی سایر آلل‌ها اختلاف آماری معنا‌داری بین دو گروه مورد و شاهد نداشتند (0/05 > P). نتیجه‌گیری: لکوس HLA-DRB1&lrm; می‌‌تواند در بیماری‌زایی پریودونتیت نقش داشته باشد. انجام مطالعه‌های مشابه با جامعه آماری بزرگ‌تر و استفاده از روش‌های تایپ کردن &rlm;HLA&rlm; با &rlm;رزولوشن بالاتر می‌‌تواند به روشن شدن مکانسیم‌های دخیل در &rlm;بیماری زایی پریودونتیت کمک کند.</div> <div style="text-align: justify;">Background and Aim: Periodontitis is one of the periodontal inflammatory diseases &lrm;and is the most &lrm;common cause of tooth loss. Although bacterial infections play a major role in the &lrm;pathobiology of periodontitis, the severity of these diseases is determined by the severity of the &lrm;host's immune response to these infections. Human leukocyte antigens (HLAs) play roles in various &lrm;biological processes, including histocompatibility, inflammation, regulation of immune response, and &lrm;the complement cascade. In the following study, we investigated the difference in the frequency of HLA &lrm;locus alleles in patients with periodontitis compared to healthy controls. Methods: This is a case- control study. Tissue samples were taken from 40 patients with periodontitis and 40 healthy controls. Case and control samples were collected based on the inclusion and exclusion criteria of the study. HLA-A, HLA-B, and HLA-DRB1 loci were genotyped by the SSP method and Olerup company kit. Allelic frequency was determined by the direct counting method, and multinomial regression models were used for statistical analysis between the two groups. Arlequin software package, version 3.5.2.2 (Excoffier and Lischer, 2010), and R software, version 3.6.1, were used to perform statistical analyses. Chi-square and OR statistical tests were also calculated. Results: This study was conducted on 40 cases and 40 controls. The two &lrm;groups were similar. The frequency of the DRB1*04 allele in the cases (28 cases) was significantly higher than the controls (16 controls) (P < 0.01). The odd ratio for this allele was equal to 2.3 (OR = 2.3). The frequency of other alleles had no statistically significant difference between the case and control groups (P > 0.05). Conclusion: HLA- DRB1 locus could play a role in the pathogenesis of periodontitis.
