fa بررسی وجود ژن mcr-1 (ژن مقاومت به کلیستین) در اشریشیا کلی جداشده از جوجه‌های یک‌روزه گوشتی نژاد راس و آرین Investigation of mcr-1 Gene (Colistin Resistance Gene) Existence in Escherichia coli Isolates Separated from Ross and Aryan Broiler Chicks میکروب‌شناسی Microbiology پژوهشی Original سابقه و هدف: اشریشیا کلی باسیل میله‌ای شکلِ گرم منفی است که به‌عنوان یک پاتوژن اصلی با طیف گسترده‌ای از اهمیت در طیور پرورشی است و در دنیا سالانه ضررهای اقتصادی قابل‌توجهی را به صنعت طیور کشور به دلیل افزایش تلفات تحمیل می‌کند. مقاومت آنتی‌بیوتیکی در این ارگانیسم به‌عنوان یک چالش اساسی در سراسر جهان مورد بحث است. بنابراین، هدف از این مطالعه بررسی وجود ژن mcr-1 (ژن مقاومت به کلیستین) در اشریشیا کلی جداشده از جوجه‌های یک‌روزه گوشتی نژاد راس و آرین است. روش کار: در این مطالعه توصیفی - مقطعی که در یک بازه زمانی چهار ماهه انجام شد، تعداد 120 جدایه غیرتکراری از اشریشیا کلی از نمونه جوجه یک‌روزه گوشتی نژاد راس و آرین که از مرغداری‌های مختلف به آزمایشگاه کوثر تهران ارسال می‌شود، جمع‌آوری شد. سویه‌های اشریشیا کلی با استفاده از آزمون‌های رایج بیوشیمیایی و میکروبیولوژی استاندارد شناسایی شدند. تعیین حداقل غلظت مهاری (MIC) با روش Colistin Broth Disk Elution طبق دستورالعمل CLSI برای آنتی‌بیوتیک کلیستین انجام شد. با روش PCR حضور ژن‌های مقاومت به کلیستین بررسی شد. یافته‌ها: 120 ایزوله اشریشیا کلی بر اساس نتایج MIC هیچ‌گونه مقاومتی به دیسک کلیستین نشان نداد و تمام لوله‌ها به جز شاهد شفاف شد. در تست پی سی آر وجود ژن mcr-1 صفر درصد گزارش شد. نتیجه‌گیری: نتیجه‌گیری می‌شود که با وجود نقش مهم حیوانات مولد غذا در انتقال مقاومت آنتی‌بیوتیکی، طیور بررسی شده منبع اصلی حمل ژن mcr-1 در ایران نبودند. این مطالعه نشان می‌دهد که بر اساس دستورالعمل‌های CLSI، روش انتشار دیسک نمی‌تواند مقاومت به کلیستین یا نگه‌داشتن عناصر ژنتیکی متحرک را منعکس کند، که برخی از مطالعه‌های قبلی در مورد مقاومت به کلیستین در ایران ممکن است گمراه‌کننده باشند. مطالعه‌های کمی با روش‌های مولکولی مقاومت به کلیستین را در ایزوله‌های اشریشیا کلی جداشده از حیوانات در ایران بررسی کرده‌اند. ارتباط احتمالی بین صنعت پرورش خوک و سطح بالای حمل mcr در برخی کشورها باید در مطالعه‌های آینده‌نگر بعدی روشن شود.   Background and Aim: Escherichia coli (E. coli) is a Gram - negative rod - shaped bacillus, which is a major pathogen with breeding damage in poultry and causes significant economic losses in the industry of Iranevery year. Antibiotic resistance in this organism is discussed as a major challenge worldwide. Therefore, the aim of this study was to investigate the presence of mcr-1 gene (Colistin resistance gene) in Escherichia coli isolates collectedfrom day - old broilers of Ras and Arian breeds. Methods: This cross - sectional descriptive study was conducted in a period of 4 months,and 120 non - repeated isolates of E. coli were collected from the samples of day - old broiler chickens of the Ras and Arin breeds. Samples were sent to the Kowsar laboratory.  E.coli strains were identified by using the Standard biochemical and microbiology routinetests = Antibiotic sensitivity test was performed according to CLSI instructions on Mueller Hinton agar medium by disk diffusion method and determination of minimum inhibitory concentration (MIC) of Colistin Broth Disk Elution was performed according to CLSI instructions for colistin antibiotic. The presence of colistin resistance genes were investigated by PCR. Results: Total of 120 E. coli isolates were collected from day - old chicks. Our results could not confirm the presence of mcr-1 positive E. coli among the studied isolates. Conclusion: It is concluded that despite the important role of food - producing animals in the transmission of antibiotic resistance, they were not the main source of mcr-1 transmission in Iran. This study showed that other mcr species (mcr-2 to mcr-9) may be responsible for the development of colistin resistance in animal isolates in Iran.
