fa اثر نانوکامپوزیت‌های مالاکیت/ اکسید روی/ کیتوزان بر تشکیل بیوفیلم و بیان ژن‌های rhlI و lasI در سودوموناس آئروژینوزا و ژن‌های fnbA و fnbB در استافیلوکوکوس اورئوس Effects of Malachite/ ZnO / Chitosan Nanocomposites on Biofilm Formation and the Expression of rhlI and lasI Genes in Pseudomonas Aeruginosa and of fnbA and fnbB Genes in Staphylococcus Aureus میکروب‌شناسی Microbiology پژوهشی Original <div style="text-align: justify;">سابقه و هدف: باکتریها، بیوفیلمها را به عنوان مکانیسمی برای افزایش بقا ایجاد میکنند و استفاده از آنتی‌بیوتیکها برای مبارزه با آنها سبب ایجاد مقاومت آنتیبیوتیکی میشود. بنابراین نیاز به تولید مواد ضد بیوفیلمی قوی برای مبارزه با بیوفیلمها است. این مطالعه تجربی با هدف بررسی اثر نانوکامپوزیتهای مالاکیت/ اکسید روی/ کیتوزان بر بیان ژنهای بیماریزایی در سودوموناس آئروژینوزا و استافیلوکوکوس اورئوس انجام شد. روش کار: در این مطالعه تجربی آزمایشگاهی نانوکامپوزیتهای مالاکیت، مالاکیت/ اکسید روی و مالاکیت/ اکسید روی/ کیتوزان تهیه شدند. میزان تشکیل بیوفیلم باکتریها با کمک رنگ کریستاله ویوله و TTC بررسی شد. حداقل غلظت مهار‌کننده تشکیل بیوفیلم و حداقل غلظت حذف‌کننده بیوفیلم تشکیلشده، برای سنجش فعالیت ضد بیوفیلمی نانوکامپوزیتها بررسی شد. میزان بقای سلولی در مجاورت نانوکامپوزیت‌ها  با رنگ‌آمیزی calcein-AM بررسی شد. بیان ژنهای rhlI و lasI در سودوموناس آئروژینوزا و ژنهای fnbA و fnbB در استافیلوکوکوس اورئوس با استفاده از تکنیکReal- Time PCR  بررسی شدند. دادهها با استفاده از آزمون آنووا تحلیل شدند. یافته‌ها: بیشترین میزان فعالیت ضد بیوفیلمی به ترتیب در نانوکامپوزیت‌های مالاکیت/ اکسید روی/ کیتوزان، مالاکیت/ اکسید روی و مالاکیت مشاهده شد. نتایج نشان داد که نانوکامپوزیت نهایی مالاکیت/ اکسید روی/ کیتوزان با غلظت  μg/ml7/81 توانست تشکیل بیوفیلم دو باکتری را حدود 10 ± 50% مهار (MBIC50) و با غلظت  μg/ml15/62بیوفیلم تشکیلشده هر دو باکتری را حدود 10 ± 50% حذف کند (MBEC50). نانوکامپوزیتهای مالاکیت / اکسید روی و مالاکیت / اکسید روی / کیتوزان بیان ژنهای rhlI و lasI سودوموناس آئروژینوزا و ژنهای fnbA و fnbB باکتری استافیلوکوکوس اورئوس را کاهش دادند (0/001 > P). استفاده ازنانوکامپوزیت نهایی مالاکیت / اکسید روی / کیتوزان 166/66 برابر بیان ژن rhlI، 30/30 برابر بیان ژنهای lasI ، 20/70برابر بیان fnbA و 100 برابر بیان  fnbBرا در مقایسه با گروه کنترل کاهش دادند (0/001 > P). نتیجه‌گیری: نانوکامپوزیتهای مالاکیت / اکسید روی و مالاکیت / اکسید روی / کیتوزان احتمالاً دارای فعالیت ضد بیوفیلمی روی باکتریهای سودوموناس آئروژینوزا و استافیلوکوکوس اورئوس هستند.  </div> <div dir="rtl" style="text-align: justify;"></div> <div style="text-align: justify;">Background and Aim: Bacteria create biofilms as a mechanism to increase survival. The use of antibiotics to fight against them causes antibiotic resistance. Therefore, there is a need to produce strong anti- biofilm substances to fight biofilms. This experimental study was conducted with the aim of investigating the effects of malachite / zinc oxide / chitosan nanocomposites on the expression of pathogenic genes in Pseudomonas aeruginosa and Staphylococcus aureus. Methods: Experimental, malachite, malachite / zinc oxide and malachite / zinc oxide / chitosan nanocomposites were prepared. The amount of biofilm formation of bacteria was checked with the help of crystal violet dye and TTC. The Minimum Biofilm Inhibitory Concentration and the Minimum Biofilm Eradication Concentration were investigated to measure the anti- biofilm activity of nanocomposites. The extent of cell survival in the vicinity of nanocomposites was evaluated by calcein-AM staining. The expression of rhlI and lasI genes in Pseudomonas aeruginosa and fnbA and fnbB genes in Staphylococcus aureus were investigated using Real- Time PCR technique. Data were analyzed using ANOVA test. Results: Based on the results of this study, the highest amount of anti- biofilm activity was observed in malachite / zinc oxide / chitosan, malachite / zinc oxide, and malachite nanocomposites, respectively. The results showed that the final malachite / zinc oxide / chitosan nanocomposite with a concentration of 7.81 μg/ml could inhibit the biofilm formation of two bacteria by about 50 ± 10% (MBIC50) and with a concentration of 15.62 μg/ml, the biofilm formed by both bacteria were remove about 50 ± 10% (MBEC50). Malachite / zinc oxide and malachite / zinc oxide / chitosan nanocomposites reduced the expression of rhlI and lasI genes of Pseudomonas aeruginosa and fnbA and fnbB genes of Staphylococcus aureus bacteria (P < 0.001). The use of malachite / zinc oxide / chitosan final nanocomposite reduced rhlI gene expression by 166.66 times, lasI gene expression by 30.30 times, fnbA expression by 20.70 times, and fnbB expression by 100 times compared to the control group (P < 0.001).
