fa مطالعه مولکولی و کلونینگ ژن dt کورینه باکتریوم در سلول مستعد برای تهیه واکسن Molecular Study and Cloning of the dt Gene of Corynebacterium in Susceptible Cells in Order to Prepare a Vaccine میکروب‌شناسی Microbiology پژوهشی Original سابقه و هدف: کورینه باکتریوم دیفتریه باکتری هوازی، گرم مثبت و غیر اسپورزا است. توکسین دیفتری (dt)، پروتئینی با سمیت بسیار بالا بوده و در اثر آلودگی باکتری به کورینه فاژ خاصی که حامل tox است، تولید می‌شود. هدف از مطالعه حاضر، کلونینگ ژن dt کورینه باکتریوم در سلول مستعدE.coli XL1blue برای تولید پروتئین نوترکیب و استفاده از این پروتئین در تحقیق‌های بعدی در سال 1401 در مؤسسه پژوهشی پاسارگارد بود. روش کار: در این مطالعه مقطعی، یک نمونه DNA سویه واکسینال کورینه باکتریوم دیفتریه 8PW تحت سویه 2000CN از موسسه رازی تهران تهیه شد. برای شناسایی ژن dt، DNA سویه باکتری با استفاده از پرایمرهای اختصاصی در روش واکنش زنجیره‌ای پلیمر از   (PCR) تکثیر یافت. با استفاده از کیت TA-کلونینگ ژن مذکور در سلول مستعد E.coli XL1blue  انجام شد. رسم درخت فیلوژنی با استفاده از تعیین توالی ناحیه ژنی srRNA  16 برای سویه باکتری استفاده شده با نرم‌‌افزارclustalX   و  Mega5  انجام شد. یافته‌ها: DNA سویه کورینه باکتریوم دیفتری حامل ژن dt بودند. انجام TA-کلونینگ ژن dt با روش‌های Real Time PCR، تعیین توالی محصول PCR، حضور کلنی‌های سفید آبی و تکثیر با پرایمر M13  تأیید شد. نتیجه تعیین توالی ناحیه ژنی SrRNA 16 تأیید‌کننده کورینه باکتریوم دیفتری بود. نتیجه‌گیری: در این بررسی ژن dt  در وکتور E.coli XL1blue  کلون و سپس تأیید شد و در جهت آمادهسازی برای ابزار پروتئین نوترکیب و همچنین به عنوان یک آنتی‌ژن قوی در تحریک سیستم ایمنی میتواند در نظر گرفته شود. این ژن می‌تواند در مطالعه‌های بعدی استفاده شود. <div> <div> <div class="msocomtxt" id="_com_1" language="JavaScript" on=""><illegal tag=""></illegal></div> </div> </div> Background and Aim: Corynebacterium diphtheriae is an aerobic, gram-positive, non- sporulating bacterium. Diphtheria toxin (dt) is a highly toxic protein and produced as a result of bacterial infection with a special corynephage that carries tox. The aim of the present study was to clone the dt gene of Corynebacterium in susceptible E.coli XL1blue cell in order to produce a recombinant protein and use this protein in subsequent research in 2022 at Pasargård Research Institute. Methods: In this Cross- sectional study, a DNA sample of Corynebacterium diphtheria vaccinal strain PW8 Sub- strain 2000CN was prepared from Razi Institute in Tehran. To identify the dt gene, the DNA bacterial strain was amplified using specific primers in the polymerase chain reaction (PCR) method. Using the TA- cloning kit, the dt gene was cloned in the susceptible E.coli XL1blue cell. The phylogeny tree for the bacterial strain was performed by DNA sequencing of the 16 srRNA gene region using clustalX and Mega5 software. Results: The DNA of Corynebacterium diphtheria carried the dt gene. TA- cloning of the dt gene was confirmed by the Real- Time PCR method, DNA sequencing of the PCR product, the presence of white- blue colonies, and amplification with an M13 primer. The result of DNA sequencing of the 16 srRNA gene region was confirmed as Corynebacterium diphtheria. Conclusion: In this study, the dt gene was cloned in the E.coli XL1blue vector and then confirmed, and it can be considered as a powerful antigen for the preparation of the recombinant protein tool, as well as for stimulating the immune system.
