fa بررسی تأثیر عصاره هیدرو‌الکلی گیاه بن‌سرخ بر شاخص‌های هومورال و سلولار سیستم ایمنی مدل موشی تحریک‌شده با ایمونوژن گلبول قرمز گوسفندی در مقایسه با داروی لوامیزول Investigating the Effects of Hydroalcoholic Extract of the Allium Jesdianum on Humoral and Cellular Indicators of the Immune System in a Mouse Model Stimulated with Sheep Erythrocyte Immunogen Compared to Levamisole سم شناسی Toxicology پژوهشی Original <div style="text-align: justify;">سابقه و هدف: سیستم ایمنی مسئول محافظت بدن نسبت به عوامل بیگانه و سرطان‌زا است و ضـعف آن می‌تواند زمینه‌ساز مرگ شود. جنس آلیاسه به‌عنوان یک کاندیدای مناسب برای حفظ هوموستئاز سیستم ایمنی مطرح‌ شده است. ازاین‌رو در این مطالعه ما به بررسی عصاره هیدرو‌الکلی گیاه بن‌سرخ (Allium jesdianum) بر پاسخ‌های مختلف سیستم ایمنی در موش پرداختیم. روش کار: در این مطالعه‌ تجربی، ما در چهار گروه 25تایی موش به بررسی اثر دوزهای مختلف (mg/kg 50، 100 و 200) عصاره‌ هیدروالکلی گیاه بن‌سرخ بر سیستم ایمنی موش پرداختیم. به این منظور تغییر وزن حیوانات، وزن اندام‌ طحال و وزن نسبی آنها، لنفوسیت‌ها خونی، قدرت تولید آنتی‌بادی به روش هماگلوتیناسیون، پاسخ ازدیاد حساسیت تأخیری، قدرت میتوژنتیسه لنفوسیت‌ها با استفاده از روش 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) بررسی شد. برای محاسبات آماری از برنامه ‌نرم‌افزاری GraphPad Prism v9.5.0.730 x64 + v8.4.0.671 استفاده شد. مقایسه بین گروه‌های مختلف با استفاده از تست‌ANOVA یک‌طرفه وPosttest Tukey-Kramer  انجام گرفت و 05/0 > p به‌عنوان تفاوت معنا‌دار در نظر گرفته شد. یافته‌ها: نتایج حاصل از این مطالعه نشان می‌دهد که بن‌سرخ نتوانست تأثیری بر جمعیت لنفوسیت‌های خونی (0/05< p) و وزن عمومی (0/05< p) داشته باشد، اما وزن طحال (22/0±  23/0) و وزن نسبی طحال (090/0±  96/0) افزایش داد. مواجه با بن‌سرخ سبب افزایش چشمگیری در پاسخ‌های تست ازدیاد حساسیت تأخیری پس از 24 ساعت (27/8±  47/53) و 48 ساعت (34/11±  81/68) نسبت به گروه کنترل شد. همچنین بن‌سرخ به‌صورت معناداری سبب افزایش تیتر آنتی‌بادی در مقایسه با گروه کنترل شد (17/0±  91/6). علاوه بر این، بن‌سرخ توانست تکثیر لنفوسیت‌های طحال را افزایش دهد (34/0±  71/7). همچنین در دوزهایmg/kg 100(09/0±  49/1) و 200 (12/0±  39/2) پاسخ تکثیر به میتوژن LPS را افزایش داد، درحالی‌که تنها در دوز mg/kg 200 (22/0±  46/2) قادر به افزایش پاسخ تکثیر به میتوژن PHA بود. نتیجه‌گیری: به نظر می‌رسد که بن‌سرخ، سبب بهبود فاکتورهای مختلف سیستم ایمنی ازجمله افزایش تکثیر لنفوسیت‌های طحال، پاسخ ازدیاد تأخیری و تولید آنتی‌بادی می‌شود. به‌طورکلی، بن‌سرخ توانست پاسخ‌های مختلف سیستم ایمنی ازجمله ایمنی همورال و ایمنی سلولی را تقویت کند.</div> <div style="text-align: justify;">Background and Aim: The immune system is responsible for protecting the body against foreign and carcinogenic agents, and its weakness can lead to death. The genus Allium has been proposed as a suitable candidate for maintaining the homeostasis of the immune system, so in this study, we investigated the effects of hydroalcoholic extract of Allium jesdianum on various immune responses in mice. Methods: In this experimental study, we investigated the effect of different doses (50, 100, and 200 mg/kg) of the hydroalcoholic extract of A. jesdianum on the immune system of 25 mice in four groups. Animal weight, spleen organ weight and their relative weight, blood lymphocytes, ability to produce antibodies by hemagglutination method, delayed hypersensitivity response, mitogenic power of lymphocytes using 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) were investigated GraphPad Prism v9.5.0.730 x64 + v8.4.0.671 software was used for statistical calculations. Comparison between different groups was done using one-way ANOVA test and Tukey-Kramer posttest, and p > 0.05 was considered as a significant difference. Results: The results of this study show that A. jesdianum could not have an effect on the population of blood lymphocytes (p > 0.05) and general weight (p > 0.05), but the weight of the spleen (0.23 ± 0.22) and the relative weight of the spleen (0.96 ± 0.09) increased. Exposure to A. jesdianum caused a significant increase in the responses of the delayed hypersensitivity test after 24 hours (53.47 ± 8.27) and 48 hours (68.81 ± 11.34) compared to the control group. Also, A. jesdianum significantly increased the antibody titer compared to the control group (6.91 ± 0.17). In addition, A. jesdianum could increase the proliferation of spleen lymphocytes (7.71 ± 0.34). Also, at the doses of 100 mg/kg (1.49 ± 0.09) and 200 mg/kg (2.39 ± 0.12), it increased the proliferation response to LPS mitogen; while only at the dose of 200 mg/kg (2.46 ± 0.22) was able to increase the proliferative response to PHA mitogen. Conclusion: It seems that A. jesdianum improves various factors of the immune system. Its effects include increasing the proliferation of spleen lymphocytes, delayed proliferative response, and antibody production.
