fa بررسی نقش انگل بلاستوسیستیس بر باکتری‌های Firmicutes و Bacteroidetes در افراد مبتلا به IBS Study of the Role of Blastocystis sp. on Firmicutes and Bacteroidetes in IBS Patients انگل شناسی Parasitology پژوهشی Original سابقه و هدف: سندرم روده تحریک پذیر (IBS) یک اختلال عملکردی شایع دستگاه گوارش (GI) است. اگرچه دلیل اصلی بیماری IBS هنوز به طور کامل مشخص نیست، ولی به نظر می‌رسد تعامل بین میکروارگانیسم‌های روده، دستگاه گوارش و سد‌ روده نقش مهمی در پاتوژنز این بیماری دارد. مطالعه حاضر با هدف بررسی اثر بلاستوسیستیس روی فراوانی Firmicutes و Bacteroidetes در بیماران IBS در مقایسه با افراد سالم انجام شد. روش کار: این مطالعه یک بررسی مورد شاهدی بود. برای این منظور، نمونه مدفوع از 80 شرکت کننده شامل 40 نمونه از هر گروه IBS و سالم بود، جمعآوری شد. هر یک از گروه‌های مذکور شامل دو زیر گروه 20 نفره (افراد دارای انگل بلاستوسیستیس  و افراد فاقد انگل بلاستوسیستیس) بودند. پس از استخراج DNA از نمونه‌های مدفوع، بررسی barcoding region  و real- time PCR کمی به ترتیب برای بررسی حضور بلاستوسیستیس  و فراوانی میکروبیوتا انجام شد. در نهایت تمامی داده&rlm;های به دست آمده از این مطالعه توسط تست‌های Shapiro- Wilk ، آزمون Student t-test، من ویتنی و ویلکاکسون بررسی شدند. یافته‌ها: در این مطالعه در بیماران مبتلا به IBS فراوانی باکتری‌  Firmicutes (3/04  ±14/52) بیشتر از گروه کنترل بود (2/19 ± 12/83). فراوانی باکتری‌ در بیماران مبتلا به IBS دارای انگل بلاستوسیستیس نسبت به گروه افراد سالم (گروه کنترل) افزایش یافته بود (3/836 ± 14/238). در مورد باکتری Bacteroidetes میزان این باکتری در افراد مبتلا به  IBS(1/69  ±8/11) بیشتر از افراد سالم ( 1/62 ± 7/62) بود. با در نظر گرفتن بلاستوسیستیس میزان این باکتری در افراد با  IBS و مبتلا به بلاستوسیستیس بیشتر از سایر گروه ها بود (1/79 ± 8/27). تغییرنسبت Firmicutes/ Bacteroidetes در گروه‌های مختلف معنا‌دار نبود. نتیجه‌گیری: در این مطالعه با وجود تغییر بین بیماران مبتلا به IBS و افراد سالم، تفاوت معنادار نبود. همچنین حضور بلاستوسیستیس تغییر معناداری روی نسبت Firmicutes/ Bacteroidetes در افراد تحت مطالعه نداشت. با این حال به نقش این تک یاخته روی باکتری‌های دستگاه گوارش باید بیشتر توجه شود. Background and Aim: Irritable bowel syndrome (IBS) is a prevalent gastrointestinal (GI) tract disorder. Although the main reason for IBS is unclear, the interaction between intestinal microorganisms and the gut barrier seems to play an important role in the pathogenesis of IBS. The current study aimed to investigate the effect of Blastocystis on the Firmicutes/ Bacteroidetes ratio of IBS patients compared to healthy subjects. Methods: This is a case- control study. Stool samples were collected from 80 participants, including 40 samples from each IBS and healthy group. Upon DNA extraction from stool samples, barcoding region and quantitative real- time polymerase chain reaction (PCR) were analyzed to investigate Blastocystis and the Firmicutes/ Bacteroidetes ratio, respectively. Student T-test, Shapiro- Wilk, Mann- Whitney, and Wilcoxon were employed to analyze data. Results: The value of Firmicutes was increased in IBS patients (14.52 ± 3.04) compared to healthy controls (12.83 ± 2.19). Regarding Blastocystis, Firmicutes were higher than other groups (14.238 ± 3.836). The value of Bacteroidetes in IBS patients (8.11 ± 1.69) was higher than healthy controls (7.62 ± 1.62). Regarding Blastocystis, the changes in IBS patients with Blastocystis were higher than other groups (8.27 ± 1.79). The changes in the Firmicutes/ Bacteroidetes ratio were not significant in different groups. Conclusion: The present study revealed that the changes in bacteria were not significant between IBS and healthy controls. In addition, Blastocystis does not change the abundance of bacteria in IBS and healthy subjects.
