fa مطالعه تغییر بیان ژن‌های Bax و Bcl-2 در رده سلولی MCF-7 تیمار شده با نانوذرات سلنیوم حاوی عصاره Anthemis atropatana The Study of Changes in the Expression of Bax and Bcl-2 Genes in MCF-7 Cell Line Treated with Selenium Nanoparticles Containing Anthemis Atropatana Extract زیست شناسی Biology پژوهشی Original سابقه و هدف: مطالعه‌ها نشان داده است که تغییر در میزان بیان برخی از ژن‌ها در ایجاد سرطان نقش دارد. هدف از این مطالعه بررسی تغییرات میزان بیان ژن‌های آپوپتوزی Bax و Bcl-2 در رده سلولی MCF-7 پس از تیمار با نانوذرات سلنیوم حاوی عصاره بابونه است. روش کار: این مطالعه از نوع تجربی مداخله‌ای است. برای انجام مطالعه‌های آماری از One way ANOVA و t-test استفاده شد. پس از سنتز نانوذرات سلنیوم حاوی عصاره <a name="_Hlk179014794">Anthemis atropatana</a>، صحت سنتز آن با SEM، TEM و EDX تأیید شد. سپس برای انجام MTT و محاسبه IC50، سلول‌های MCF-7 (رده سلولی سرطانی) و  MCF-10(رده سلولی سالم)، به مدت 24 ساعت با نانوذرات سلنیوم، تیمار شدند. در نهایت تغییرات میزان بیان ژن‌های Bax و Bcl-2 مطالعه شدند. یافته‌ها: نتایج بررسی صحت سنتز نانوذرات سلنیوم، نشان داد که نانوذرات سنتز شده کروی بوده و دارای قطر 33/40 نانومتر هستند. غلظت IC50، برابر با µg/ml 6 به دست آمد. نتایج Real time PCR نشان داد که پس از 24 ساعت تیمار در غلظت IC50 از نانوذرات سلنیوم، میزان بیان ژن آپوپتوزی Bax، 0/31 ± 2/69 برابر افزایش بیان معنادار (0/001 > P-value) و ژن ضد آپوپتوز Bcl-2 به میزان 0/17 ± 0/21برابر کاهش بیان معنادار (0/001 > P-value)  پیدا کرده است. نتیجه‌گیری: نانوذرات سلنیوم حاوی عصاره بابونه توانستند بیان ژن‌های درگیر را به نفع القای آپوپتوز تغییر دهند.   <pre style="text-align:justify; margin-bottom:11px"> Abstract Background and Aim: Studies have shown that changes in the expression level of some genes play a role in the development of cancer. The aim of this study is to investigate the changes in the expression of apoptotic genes Bax and Bcl-2 in MCF-7 cell line after treatment with selenium nanoparticles containing chamomile extract. Methods: This study is a practical and experimental intervention. One- way analysis (ANOVA) and t-test was used for statistical analysis. After synthesizing selenium nanoparticles with Anthemis atropatana extract, the correctness of the synthesis was confirmed by SEM, TEM and EDX. To perform MTT and calculate the IC50, the cells (cancer cell line) MCF-7 and (healthy cell line) MCF-10 were treated with selenium nanoparticles for 24 hours. Finally, the changes in Bax and Bcl-2 gene expression were examined. Results: The results of checking the accuracy of the synthesis of selenium nanoparticles showed that the synthesized nanoparticles were spherical and had a diameter of 33.40 nm. The IC50 concentration was 6 µg/ ml. The results of real- time PCR showed that after 24-hour treatment with the IC50 concentration of selenium nanoparticles, the expression of apoptotic gene Bax increased significantly by 2.69 ± 0.31-fold (P-value < 0.001) and the expression of anti- apoptotic gene Bcl-2 decreased significantly by 0.21 ± 0.17-fold (P-value < 0.001).
