fa تمایز انتاموبا هیستولیتیکا و انتاموبا دیسپار با یک واکنش زنجیره‌ای پلی‌مراز آسیب‌شناسی Pathology پژوهشی Original سابقه و هدف: با اینکه تشخیص میکروسکوپی قادر نیست تک‌یاخته‌های مشابه نظیر انتاموبا هیستولیتیکا/ انتاموبا دیسپار را از یکدیگر متمایز نماید، ولی هنوز تشخیص آمیبیازیس، مبتنی بر روش‌های میکروسکوپی است. لذا نیاز مبرمی برای ابداع و راه‌اندازی یک تکنیک ساده، ارزان و قابل اجرا در آزمایشگاه‌های تشخیص طبی به منظور شناسایی و افتراق این دو گونه وجود دارد. روش بررسی: به منظور تعیین گونه ایزوله آمیب موجود در نمونه‌های مشکوک، اقدام به بررسی ملکولی به روش واکنش زنجیره‌ای پلی‌مراز (PCR) گردید. برای افتراق همزمان این دو گونه، از یک جفت پرایمر pEd21/30 مربوط به ژن Peroxiredoxinاستفاده شد. با انجام PCR بر روی DNA استخراج شده موارد مثبت انتاموبا هیستولیتیکا با این پرایمر باند زیر 100 جفت باز را نشان داده و موارد مثبت انتاموبا دیسپار قطعه ای بالای 100 جفــت باز را تکثیر نمود. یافته‌ها: دراین مطالعه از 22 نمونه مثبت از نظر میکروسکوپی، در یک بیمار انتاموبا هیستولیتیکا مشاهده شد و بقیه نمونه‌ها (21 نمونه)، همگی از نظر انتامبا دیسپار مثبت بودند. نتیجه‌گیری: به نظر می‌رسد که با یک جفت پرایمر در تکنیک PCR می‌توان انتاموبا هیستولیتیکا و انتامبا دیسپار را تکثیر نمود که این روشی آسان‌تر و مقرون به صرفه جهت تشخیص و تمایز این دو گونه می‌باشد. واژگان کلیدی: انتاموبا هیستولیتیکا، انتاموبا دیسپار، ژن Peroxiredoxin Abstract: Background and Aim: In most part of the world detection of cysts and trophozoites of Entamoeba is based on morphological structure of this species in stool sample by microscopy. However, microscopic examination is unable to distinguish between similar morphological protozoa such as Entamoeba histolytica and Entamoeba dispar. A simple and cost-effective method is needed in medical laboratories for detection and differentiation of these two species. Materials and Methods: Stool samples of patients who were referred from health care centers were examined by direct microscopy and trichrome stain. Polymerase chain reaction (PCR) utilizing *pEd30F and pEd21AS primers from Peroxiredoxin gene, was used for differentiation of E. histolytica and E. dispar. Genomic DNA from samples was amplified by these primers. The fragment under 100 bp was related to E. histolytica and in contrast the fragment above the 100 bp was related to E. dispar. Results: In this study from 22 microscopic positive samples, E. histolytica was observed only in one patient and E. dispar was detected in the other 21 samples. Discussion: The result of this study indicate that the PCR reaction could amplify E. dispar and E. histolytica with just one primer pair and this is a cost-effective method for distinguishing between these two species.
