Background: Choroid plexus (CP) is a branched structure made up of a single layer of epithelial cells and blood capillaries, forming the blood-CSF-barrier. The CSF (cerebrospinal fluid) is mainly produced from the CP. Aquaporin1 (AQP1), water channels that are highly expressed on the apical side of the membrane in choroid plexus, have a major role in mediating water transport across the apical membrane of choroid plexus during CSF secretion. The AQ1 expression alters in many CNS diseases, so determination of the exact site as well as the amount of AQ1 expression in CP cells is essential.
Materials and methods: Choroid epithelial cells of lateral ventricle in Wistar rats were isolated and after mechanical and enzymatic digestion, grown in DMEM supplement. Flow cytometry and Western analysis were used for detecting AQP1 expression in the rat CP epithelium and immunocytochemistry was done for localization of AQP1.
Results: Western blotting detected peptides with molecular masses of 27 and 32 kDa for AQP1 antibody in the choroid plexus of the fourth ventricle. Immunocytochemistry demonstrated that AQP1 is expressed in the cytoplasmic membrane of these cells. Also flow cytometry showed that 97.63% of choroid epithelial cells express AQP1.
Conclusion: This study demonstrates high expression of AQP1 in choroid plexus cells and shows that AQP1 expression is restricted to the cytoplasmic membrane.
